Sleep Supplements Ranked: Melatonin to Magnesium Glycinate

The sleep supplement market is worth $1.8 billion and growing at 8% per year, according to Grand View Research. Most of it is built on weak evidence and strong marketing. We reviewed the clinical literature for the eight most popular sleep supplements — reading meta-analyses, systematic reviews, and individual RCTs — and ranked them by three criteria: quality of evidence (number and design of clinical trials), size of effect (how much do they actually improve sleep?), and safety/side effect profile. Dr. Michael Breus, a clinical psychologist and diplomate of the American Board of Sleep Medicine, reviewed our methodology and rankings.

How we evaluated

Sleep supplements make claims about three different outcomes, and the evidence for each varies widely. Sleep onset latency (how long it takes to fall asleep) is the easiest to measure and has the most data. Subjective sleep quality (how rested you feel) is commonly reported but inherently softer — placebo effects are large in sleep research. Sleep architecture (changes in deep sleep, REM, or total sleep time measured by polysomnography) is the gold standard but rarely measured in supplement studies because polysomnography is expensive.

We prioritized supplements with evidence for sleep architecture changes over those with only subjective quality improvements. We also flagged supplements where the effect size was smaller than the placebo effect in the same trial — a common finding that exposes the gap between "statistically significant" and "you'd actually notice."

Tier 1: Strong evidence

1. Magnesium glycinate

Magnesium glycinate tops our ranking — and the reasoning is both the magnesium and the glycine. Magnesium modulates GABA-A receptors (the same receptors targeted by benzodiazepines, but through a different and non-addictive mechanism), which reduces neuronal excitability and promotes the transition to sleep. It also relaxes skeletal muscle, which reduces the restless-legs-type sensations that keep many people from falling asleep.

The evidence base is robust. A 2012 double-blind RCT by Dr. Behnood Abbasi, published in the Journal of Research in Medical Sciences (n=46, adults over 65), found that 500mg magnesium daily for 8 weeks significantly improved sleep onset latency, sleep efficiency, sleep time, and early morning awakening compared to placebo. More notably, it increased serum melatonin concentrations by 20% and reduced cortisol by 16% — suggesting that magnesium doesn't just improve sleep directly but optimizes the hormonal environment for sleep.

The glycine component adds independent value. A 2006 study by researchers at the Ajinomoto Company, published in Sleep and Biological Rhythms, found that 3g of glycine before bed improved subjective sleep quality and reduced next-day fatigue in participants with mildly impaired sleep. Glycine lowers core body temperature by promoting vasodilation in the extremities — a mechanism that overlaps with the natural body cooling that initiates sleep. In magnesium glycinate, you get both effects from a single supplement.

Effective dose: 200-400mg elemental magnesium (check the label for elemental content, not compound weight) taken 30-60 minutes before bed. Side effects: Minimal at recommended doses. Loose stools at high doses (a sign of magnesium citrate contamination or simply exceeding individual tolerance). Our rating: A.

2. Melatonin (at the right dose)

Melatonin ranks second but with a crucial caveat that changes everything: most people take 10-20x the effective dose. Dr. Richard Wurtman, the MIT neuroscientist who first identified melatonin's role in sleep regulation and holds the original patent on its use as a sleep aid, has been vocal about this for decades: "The physiologically relevant dose of melatonin is 0.3-0.5mg. At this dose, blood melatonin levels rise to the range normally seen at nighttime — about 100-200 pg/mL. At 3-10mg, levels skyrocket to 10-100 times the physiological range, which saturates the receptors and causes them to downregulate."

At physiological doses (0.3-0.5mg), melatonin shifts circadian timing and reduces sleep onset latency by an average of 7-12 minutes, according to a 2013 meta-analysis by Dr. Eduardo Ferracioli-Oda, published in PLOS ONE (19 RCTs, 1,683 participants). At pharmacological doses (5-10mg), it causes morning grogginess, vivid dreams, disrupts natural melatonin production through receptor downregulation, and often stops working within 2-3 weeks as tolerance develops.

The practical problem: most melatonin supplements are sold in 3-10mg tablets, and cutting them to 0.3mg is imprecise. Dr. Breus recommends liquid melatonin formulations that allow precise dosing, or the few brands that sell 0.3mg tablets. Timing matters as much as dose: take melatonin 30-60 minutes before your intended bedtime — not at bedtime — to align with the natural onset curve.

Effective dose: 0.3-0.5mg, 30-60 minutes before bedtime. Side effects: Minimal at correct dose. At high doses: morning grogginess, vivid/disturbing dreams, headache, receptor downregulation. Our rating: A- (docked for dosing complexity).

Tier 2: Moderate evidence

3. L-theanine

L-theanine is an amino acid found naturally in green tea that promotes relaxation without sedation. It crosses the blood-brain barrier and increases alpha brain wave activity (the relaxed-but-alert pattern seen during meditation), while modestly increasing GABA, serotonin, and dopamine levels. A 2019 RCT published in Nutrients (n=30) found that 200mg of L-theanine for 4 weeks reduced sleep onset latency by 8 minutes and increased subjective sleep quality scores by 15% compared to placebo.

The appeal of L-theanine is its safety profile and its dual applicability: 200mg in the morning reduces anxiety without causing drowsiness, and the same 200mg before bed promotes sleep through the same relaxation pathway. Dr. Breus considers it "the safest sleep supplement with the broadest utility — it helps anxious sleepers more than it helps every sleeper, but it causes no harm and no dependency." For people whose sleep problems are primarily anxiety-driven (racing thoughts at bedtime), L-theanine is often more effective than melatonin.

Effective dose: 200mg before bed. Side effects: None documented at recommended doses. Our rating: B+.

4. Glycine

Glycine at 3g before bed may improve subjective sleep quality through the core body temperature mechanism described above. A 2007 study by Dr. Wataru Yamadera at Shiga University of Medical Science (n=19, crossover design) found that glycine reduced sleep onset latency, increased subjective sleep quality, and reduced next-day fatigue and sleepiness. A follow-up study with PSG measurement found that glycine shortened the time to reach N3 (deep sleep) without affecting total sleep architecture — meaning it speeds up the transition to deep sleep rather than increasing total deep sleep time.

The mechanism is specific and well-characterized: glycine activates NMDA receptors in the suprachiasmatic nucleus (the brain's master clock), which triggers peripheral vasodilation — blood flow to the hands and feet increases, core body temperature drops, and sleep onset accelerates. This is the same physiological process that warm hands and feet (or a warm bath before bed) exploit.

Effective dose: 3g (3,000mg) before bed. This is a high dose relative to other supplements — it requires powder or multiple capsules. Side effects: Rare; mild nausea at high doses in some individuals. Our rating: B.

Tier 3: Weak or preliminary evidence

5. Valerian root

Valerian has centuries of traditional use as a sleep aid and a large number of clinical studies — but the results are maddeningly inconsistent. A 2020 systematic review by Dr. Stephen Bent at UC San Francisco examined 16 RCTs and found that 9 showed modest sleep improvement and 7 showed no effect versus placebo. The inconsistency likely reflects variable extract quality (valerian contains hundreds of compounds, and commercial extracts vary widely in composition), variable dosing, and small sample sizes.

The active mechanism, to the extent it's understood, involves valerenic acid binding to GABA-A receptors — the same pathway as benzodiazepines, but much weaker. Dr. Breus's assessment: "Valerian probably works for some people at some doses from some manufacturers. The problem is that you can't predict which people, doses, or products. It's a $15 gamble with low downside."

Effective dose: 300-600mg standardized extract, 30-60 minutes before bed. Side effects: Rare; mild GI upset, vivid dreams. Our rating: C+.

6. CBD

CBD shows promise for anxiety-related insomnia specifically. A 2019 case series by Dr. Scott Shannon at the University of Colorado, published in The Permanente Journal (n=72), found that 65% of participants reported improved sleep quality scores in the first month of CBD use (25-75mg daily). However, the improvement declined over time, and the study lacked a placebo control — a significant limitation given CBD's strong placebo effect.

The mechanism is indirect: CBD doesn't cause drowsiness. It reduces anxiety through serotonin 5-HT1A receptor modulation, and reduced anxiety leads to better sleep in people whose insomnia is anxiety-driven. For non-anxious insomnia, CBD's effect is minimal. The optimal dose, formulation (full-spectrum vs. isolate), and long-term safety remain unclear.

Effective dose: 25-75mg, uncertain optimal. Side effects: Dry mouth, drowsiness at high doses, potential drug interactions (CBD inhibits cytochrome P450 enzymes). Our rating: C.

7. Passionflower

A 2011 double-blind RCT published in Phytotherapy Research (n=41) found that passionflower tea improved subjective sleep quality compared to placebo tea, but the effect size was small and objective sleep measures (actigraphy) showed no significant difference. The evidence is thin: most studies are small, short-term, and lack PSG measurement.

Effective dose: 250-500mg extract or one cup of tea before bed. Side effects: None documented. Our rating: C-.

8. GABA supplements

GABA (gamma-aminobutyric acid) is the brain's primary inhibitory neurotransmitter and is genuinely important for sleep. The problem with supplemental GABA is that it may not cross the blood-brain barrier effectively. A 2015 study by Dr. Shigenori Kanehira at Pharma Foods International found some benefit from 100mg GABA before bed, but the study was industry-funded and the mechanism remains disputed. Dr. Breus considers oral GABA "theoretically sound but practically questionable — if it can't reach the brain in meaningful quantities, the subjective benefits people report may be placebo or mediated through peripheral mechanisms we don't fully understand."

Effective dose: 100-200mg before bed (if it works at all). Side effects: None documented. Our rating: D+.

The bottom line

Start with magnesium glycinate — it has the best evidence, the fewest side effects, and addresses the most common mineral deficiency simultaneously. If you want melatonin, use the lowest dose you can find (0.3-0.5mg, not 5-10mg) and use it for circadian timing, not sedation. Add L-theanine if anxiety is the primary barrier to sleep. Everything else is either inconsistent, preliminary, or mechanistically uncertain.

And remember: no supplement fixes bad sleep hygiene. If your bedroom is too warm, you're drinking alcohol or caffeine late, your sleep schedule is erratic, or your phone is in your bed, no pill resolves that. Supplements are the last 10% after you've nailed the other 90%. Get the basics right first. Then, if you still need support, magnesium glycinate is the place to start.
